Meaning

"Back to the grind" means returning to your normal routine of work or study after a break. "The grind" is hard, repetitive work, like a millstone turning around and around to grind grain into flour. People also talk about "the daily grind", the ordinary, tiring rhythm of every working day.

It's casual, and it's usually said with a sigh and a smile. You say it on a Sunday night, on the first morning after a vacation, or at the end of a long weekend: "Well, back to the grind." It complains a little, but mostly it's a shared joke between people who all have to go back.

It's fine with coworkers and friends. Because it jokes that your job is dull, don't use it in a job interview or to a manager who just gave you an exciting new project. You'll also hear "back to reality", which means the same thing and sounds a bit softer.
