Meaning
To call it a day is to stop working for now, usually because it is late, you are tired, or you have done enough. The picture is simple: you look at what you have done and decide it counts as a day's work. It is neutral and friendly, and it is almost always said as a suggestion: "let's call it a day".
There is a second, bigger meaning: to stop something for good. "After twenty years, the band called it a day." Context tells you which one is meant; in an office at six o'clock it is the small one.
